Kenji Yamauchi founded Yamanouchi Pharmaceutical Trading Company in Osaka, and in 1940, the company was renamed Yamanouchi Pharmaceutical Co., Ltd.

Astellas was established in April 2005 through the merger of two companies – Yamanouchi Pharmaceutical (founded in 1923) and Fujisawa Pharmaceutical (founded in 1894).
